will my son lose his playlist if i click new playlist on itunes

I have just got an iphone 5S. I want to put some tunes on. I went on iTunes, which my son has been using for a while, and found he had a lot on the playlist. In the top corner I noted an icon which when clicked showed "new playlist". if I click this will my sons playlist be deleted.

No, you can setup multiple playlists. It's safe to add another.
