Is it possible to put a folder of apps into another folder of apps?

Nope. Folders can't be put into other folders.

Can it be done on the iPhone 4s? Because I'm sure my brother done it to my mums iPhone

Depending on your version of iOS, it can kind of be done, but it's just a glitch. If you actually try to use those inner folders, your phone will crash (however, you should be able to reach them via Spotlight without issue). See here:
