Need to know how to configure 2017 27" iMac 5k retina
There is a small 28 gb drive that works fine for a basic install but when I add Apps there is not much space.
I have manaully moved new apps to the 1tb drive and that works fine.

Is there a method of making the default download directory on the 1tb

Your post is not quite informative enough for a definitive response, in my opinion. With that being said, which of the two drives you mentioned the internal boot drive? If the 1TB drive an external drive then you can install macOS onto it and then boot from it, if necessary. The link below will provide instructions for you.
