Does number change to iPhone 4 on activation or do you have to go into a brick and mortar location to transfer number??

If you're replacing an existing phone then activating the iP4 is no different than activating any other phone, with the exception that you have to use iTunes to do it.

I transferred number over the phone with csr. It only took a few minutes if that's what your asking. I activated the phones via iTunes on the numbers they came with then called and and swapped numbers so the iPhones were on the lines I wanted.
