Are you running a mail server on your computer? If you're simply trying to access email from your your ISP, Giogle, Yahoo, Windows Live, etcetera, all you have to do is go to Settings/Mail, Contacts, Calendar on your iPhone and then select "Add Account". Follow the simple directions on the screen.
