On new iPhone6 I did not check "icloud Photo Library". I get a warning when turning it on about removing the photos from iTunes. I want to make sure if I click remove, it will not remove them from the phone, so I can then back them off to the iCloud.

Enabling iCloud Photo Library will NOT remove any pics from your device. It will sync the pics to the rest of your same configured devices, if you have any. Note: with Photo stream and iCloud Photo Library, what you do on one device will be reflected on the other(S). If you disable iCloud Photo Library, the pics on the device will remain intact, but you will lose the photo syncing ability to that particular device.

Keep in mind that iCloud is not like Dropbox. It's not a storage solution. It's a back up option. So if you back your photos up to iCloud or use iCloud Photo Library, then delete the photos off your camera roll, then they also delete from iCloud.
