The whole iTunes music folder. Just look for the path to that folder in the iTunes preferences window.

If you want pretty much everything to stay the same, make sure you back up not just the iTunes Media folder (if that's where you keep your media), but make sure you backup your whole iTunes folder (which is in your music folder by default). The "iTunes Library Extras.itdb", "iTunes Library Genius.itdb", "iTunes Library.itl", and "iTunes Music Library.xml" are what store what songs you had in iTunes and where they were on your hard drive (try to restore them to the same path then you won't have to worry about iTunes finding them), which songs were in which playlists, your Genius mixes and information, and some other things. The only thing it doesn't back up is preferences which is easy to redo.
