Would it be best to update the os before anything else or get the phone all set up and activated first?

I'm sure it will ship already with 4.2.6 but somehow if it doesnt, just register it first on iTunes and then you can upgrade the OS at anytime

If the phones don't ship with the new OS but it is available for update, you will be prompted in iTunes AFTER it's activated. Once activated then you can update. There's no significance to updating the phone before you activate it.
