I've lost my library or my itunes wont read what i've downloaded?

I have A 64 GIG ipod 5. Running off of Windows 10. I have experienced this before when I updated the software on my Zune this same thing happened.
I am afraid to hook my ipod up to my laptop for fear of it being changed to what is now in my itunes which is a tiny fraction of what I had? can someone help me? I have 5400 songs on my laptop and about 20 playlists that now don't show up in my itunes? Should i hook my ipod up to my laptop and see if that will solve the problem or clear it up strictly on the laptop? i just dont want to lose all my music on the device since I have a trip coming up where I use the device practically all day!

You may need to connect to iTunes and sync them back to the iPod. Someone correct me if I'm wrong. My sister lost her music on her phone and had to do that. It was after updating iOS 9.
