No no, restoring from a backup will not remove the jailbreaked. Instead he need to manually do a backup via iTunes, once backup is finish, put the iPhone in recovery mode, then use iTunes to download the latest iOS and install.
The recovery mode is important as it will prevent iTunes to do some sort of checking prior in factory restore.
If the device is not in recovery mode, and you click on upgrade or restore, iTunes will still download the latest iOS and proceed to the installation, but during the installation, you will face load of errors throw at you by iTunes which will render factory restore impossible.
So manually backup in iTunes, then put device in recovery mode, then let iTunes upgrade or restore.
