You just need to either plug the iPhone into the computer and open iTunes and make a backup, or go to Settings-iCloud-Storage And Backup-Back Up Now.

No don't need to lock your phone to back it up as mentioned above you can do the iTunes route or via iCloud

For iCloud you must first enable iCloud back up and have your phone plugged in and have wifi with a strong signal

To add one thing to Katarans explanation i have a feeling and if i am right when talking about being locked... Your lock screen has to be closed and be on that screen which we would consider it being locked. It is just a matter of words and everything else he said holds true.
